Outline of Research at the Start

多くの植物病原菌の宿主特異性の成立には、エフェクターと総称される分泌タンパク質群が重要な役割を果たしていると推察されるが、その分子基盤は不明である。本研究では、先駆的に発見に成功している、宿主特異性成立に必須である病原菌エフェクターの解析を起点に、エフェクターを介した植物病原菌の宿主特異性成立の分子基盤を解明することを目的とする。さらに、本成果に基づき、エフェクターの戦略を遮断する、新たな耐病性作物の創出技術を開発する。

Outline of Annual Research Achievements

本年度は、まず、ウリ類炭疽病菌のウリ科作物への宿主特異性に関与するエフェクターEPC3について、その機能ドメインEPC3-NDの立体構造をNMR解析により決定した。さらに、決定したEPC3-NDの立体構造、および標的因子BIP5の立体構造情報をもとに、両因子の結合領域を予測し、その予測に基づき、BIP5に7アミノ酸の変異を導入した結果、EPC3-NDとBIP5の結合はキャンセルされた。一方、EPC3-NDとBIP5の結合は、植物細胞で共発現させたサンプルに対する共免疫沈降解析、および、酵母ツーハイブリッド解析で検出される一方、大腸菌で発現させたEPC3-NDを用いた場合、BIP5との結合は検出されなかった。さらに酵母細胞で発現させたEPC3-NDタンパク質を用いたアッセイでは結合が検出され、真核細胞におけるEPC3-NDの修飾などがBIP5との結合に関与する可能性が提示された。また、キュウリにおいて、エフェクターEPC3の標的であるBIP5をコードする遺伝子をウイルス誘導遺伝子サイレンシング(VIGS)によってノックダウンした結果、PAMP処理によって誘導される活性酸素生成の低下が観察された。また、EPC1の標的であるIOS1をコードする遺伝子をVIGSによってノックダウンした結果、やはりPAMP依存的活性酸素生成の低下が観察された。以上より、キュウリにおけるPAMP誘導免疫に両因子が関与することが強く示唆された。また、ウリ類炭疽病菌のウリ科作物への宿主特異性に関与する転写因子TFV1、TVL1、TVL2をコードする遺伝子破壊株の遺伝子発現解析により、新たな病原性エフェクター遺伝子候補を絞り込み、一部について遺伝子機能解析を実施し、その結果、ウリ科作物への宿主特異性に関与する新規エフェクター遺伝子EPC5およびEPC6の同定に成功した。
