Outline of Research at the Start

自閉スペクトラム症は社会的コミュニケーションの障害を特徴とする疾患であり、症状の一つに感覚の過敏性がある。私たちは自閉スペクトラム症の発症に関連するShank3というたんぱく質が、感覚神経の形成と発達に重要な役割を担っている可能性を見出した。この研究ではShank3のさらなる研究を通じて自閉スペクトラム症で感覚過敏性が起こる原因を明らかにし、治療薬の候補となるような薬剤の開発を目指している。

Outline of Final Research Achievements

This study looked into why children with autism spectrum disorder (ASD) show hypersensitivity very often. To answer to this question, we used mice that were missing an ASD-related gene, Shank3. These mice had an imbalance between excitatory and inhibitory neurons in the thalamus, a particular brain region processing the sensory inputs. We also found that the mice developed generalized seizures very easily. Shank3 protein binds to another protein that maintains the harmonized gene functions in the brain. Moreover, loss of Shank3 affects the way how the ASD-related genes are decoded. These results suggest that hypersensitivity and seizures are both linked to maturation of thalamic functions during early childhood. By deeply understanding the function of Shank3, clinically useful therapies for both symptoms can be invented in the future.

Academic Significance and Societal Importance of the Research Achievements

自閉スペクトラム症(ASD)は年々増加傾向にあり、支援の重要性が増している。ASD罹患児の90%以上に感覚過敏や鈍麻が見られる。それ自体が、偏食をはじめ、生活に大きな影響を及ぼしているだけでなく、対人関係や興味の偏り、様々な場面でのこだわりやかんしゃくにも、感覚神経の入力と出力の不具合が関与すると考えられている。またASD患者の10-20%はてんかんを合併し、知的発達の遅れが強いほどてんかんの合併が多い。このようにASDの中心症状である感覚神経の障害とてんかん、双方に共通する原因を解明することで、両症状を軽減する新しい治療法の開発が可能となる。
