Outline of Research at the Start

スキルス胃癌は診断時に腹膜転移をはじめとした遠隔転移を伴うことが多く、外科的に切除可能病変であっても再発率が高いことから、その治療成績は極めて不良である。スキルス胃癌は癌細胞の周囲間質に線維組織を豊富に形成し、その悪性度に寄与することが示唆されているがその起源は不明である。本研究では、線維芽細胞に形態が酷似する、多分化能をもつ脂肪前駆細胞に注目し、スキルス胃癌間質の線維組織形成に深く関わるのでは推察し、その検証を行う。脂肪前駆細胞からスキルス胃癌関連関連線維芽細胞形成のメカニズムを追求することによりスキルス胃癌の成り立ちの一部を解明し、スキルス胃癌に対する新規治療開発を目指す。

Outline of Annual Research Achievements

脂肪前駆細胞(PrAd)をスキルス胃癌細胞株と共培養することにより、スキルス胃癌細胞株の細胞の増殖促進傾向をみとめた。また逆に、スキルス胃癌細胞株との共培養により、PrAdの増殖も有意に促進されるとともに、前駆細胞から成熟脂肪細胞への分化能が低下した。
さらに、スキルス胃癌細胞株と共培養することにより、PrAdのDLK-1発現は低下するとともに、糖鎖修飾をおこすことがわかった。PrAdはスキルス胃癌存在下において、脂肪前駆細胞としての性質を喪失し繊維芽細胞として増殖し、さらに胃癌細胞増殖を増強させるという悪性化サイクルにかかわる可能性がある。

Strategy for Future Research Activity

切除したスキルス胃癌組織を用い、間質領域におけるDLK-1発現を免疫染色にて検討する。
スキルス胃癌細胞との共培養により、PrAd中でおきるDLK-1の糖鎖修飾を同定する。PrAdをGFP標識し、超免疫不全マウスを用い、スキルス胃癌細胞株と同時移植を行い、胃癌細胞および線維芽細胞増殖の挙動を評価する。
