Outline of Research at the Start

本研究ではミトコンドリアの未だ知られていない構造を解析することで、ミトコンドリアが増える過程を明らかにします。ミトコンドリアはかつて円筒形の構造体だと広く知られていますが、一定の条件下でミトコンドリアの膜が平たい「シート」を形成することを発見しました。この現象を顕微鏡観察や生物化学的な手法を用いて網羅的に解析し、「シート」の構造や生理的機能を明らかにします。また、生物物理学の観点から細胞内の膜動態を調査することも目標とします。

Outline of Annual Research Achievements

In the past year, I have finalised experiments in the project and additionally conducted analyses of mammalian cells. I have also prepared a manuscript that provides a detailed account of the key phenotypes outlined in the proposal for this grant. I am currently waiting for final electron tomography analyses of these samples ahead of submission of this work. I anticipate that the manuscript will be submitted within the next two months, and the paper will be accepted for publication by the end of FY2024. I have also collaborated on several projects that have related in publications or at the final stages of revision. Accepted papers include 10.1083/jcb.202306120 and 10.1038/s44318-024-00091-8 .

2: Research has progressed on the whole more than it was originally planned.

Reason

I have not finished this project as early as I had expected. However, this is due to additional important data becoming available related to similar phenomena in higher eukaryotes. For this reason, I am in the process of collecting data to show the conservation of mitochondrial membrane dynamics throughout eukaryotes. These data will result in a higher-quality publication with broader implications.

Strategy for Future Research Activity

As this is the final year of this project, I will spend the remaining time finalising the manuscript for submission and then conducting revision experiments as necessary. As it will be necessary to respond to reviewer requests, I am unable to provide a defined plan for the remainder of the project.
