Outline of Research at the Start

Outline of Final Research Achievements
Culturing mesenchymal stem cells (MSCs) on elastic gels patterned with cell-scale hard and soft regions and letting them migrate non-persistently for extended periods greatly amplifies long-period fluctuations in intracellular stress and nuclear shape, acting as a hormesis-like mechanical stimulus. This “mechano-activation” broadly up-regulates genes involved in proliferation, survival, motility, and differentiation, enabling the generation of high-function MSCs with enhanced therapeutic potential. We systematically explored pattern designs that maximize this effect and introduced the wavelet energy density of cell-area change acceleration as a new in-situ index for quantifying stress non-equilibrium. Using this metric, we determined that a triangular 3P pattern with a hard/soft modulus combination of 150 kPa/5 kPa most strongly promotes mechano-activation. These results expand the technological foundation for producing therapeutically superior MSCs.

Academic Significance and Societal Importance of the Research Achievements
