Outline of Research at the Start

辺縁皮質を対象とした深部脳刺激術(DBS)による神経応答の作用機序を明らかにするため、深部脳刺激術の神経応答を機能MRI 法によって全脳で計測する。将来的に、ヒトのうつ病、強迫性障害の治療という臨床応用につなげるためには、外科手術を必要とする現在の DBS ではなく、非侵襲で、脳の深部をターゲットとできる刺激法が望まれる。そこで、ヒトやマカクザルの脳に対する経頭蓋骨な脳刺激法として注目されている超音波を利用し、超音波刺激による DBS の実現を目指し、マカクザルの大脳辺縁系を対象として、新しい刺激法の開拓を行う。

Outline of Annual Research Achievements

独自に開発したマルチサイト記録法を駆使して、EMによって人為的な不安状態の誘導を行い、その間に、不安ネットワークから神経活動の多点同時記録を行った。こうした不安ネットワークに対して、多点同時LFP記録を行い、ベータ波を介した情報の流れがあることを新たに発見した。この研究では、辺縁系皮質と認知系皮質を含むこの4つの領野から、課題遂行中の神経活動の同時記録を行い、神経振動を介した階層的な相互作用が存在することを明らかにした。これらの大規模データをもとに、神経振動のグレンジャー因果解析、スパイク駆動 LFP解析など、神経信号の方向性を明らかにするネットワーク解析を行い、不安コネクトームを構成する4つの領野の間のトップダウン信号を同定し、Nature Communications 誌にアクセプトされた。

Reason

本年度は、マカクザルにBalloon analog risk task (BART)を訓練し、計算論モデルを導入することで、サルがリスクや予測のあいまいさに対する悲観的な価値判断を行っていることを、定量的に明らかにした。EM刺激の準備も順調で、研究はおおむね順調に進展している。

Strategy for Future Research Activity

ヒトやマカクザルの脳に対する経頭蓋骨な脳刺激法として注目されている超音波を利用し、超音波刺激による DBS の実現を目指し、マカクザルの大脳辺縁系を対象として、新しい刺激法の開拓を行う。
