Outline of Research at the Start

膵癌が治療抵抗性である要因として腫瘍免疫抑制的な環境があるが、癌間質細胞(CAF)が原因の一つであると考えられている。また、CAFのheterogeneityの解明が進み、膵癌でも複数の機能的CAFクラスターの存在が明らかにされてきたが、腫瘍免疫抑制の要因となる機能的CAFクラスターは同定されていない。本研究では抗腫瘍免疫抑制的な環境を構築する特定のCAFクラスターを同定し、そのようなCAFクラスターを標的とするキメラ抗原受容体T細胞(CAR-T)療法を開発する。

Outline of Final Research Achievements

From scRNA-seq data of 24 pancreatic cancer patients, 496 genes associated with cancer-associated fibroblasts (CAFs) and patient prognosis were identified. Among these, 130 genes were found to correlate with T cell exhaustion. Candidate genes potentially useful for CAR-T cell therapy were then selected, and their expression was confirmed via immunohistochemistry. CAR-T cells were subsequently generated. In a killing assay, cytotoxicity against CAFs was observed. Future studies will involve administering these CAR-T cells in a human pancreatic cancer mouse model.

Academic Significance and Societal Importance of the Research Achievements

我々は、膵癌特有の腫瘍免疫抑制微小環境の要因の一つである特定のCAFクラスターを標的とするため、キメラ抗原受容体T細胞(CAR-T)の手法に着目した。scRNAseqデータに基づいて特定の機能的CAFクラスターを標的とすることで、癌間質相互作用や物理的な間質構造改変のみならず、免疫抑制性微小環境の改変を実現する。それにより、固形がんに対する様々な治療薬の長期投与に必ずつきまとう免疫の疲弊化や免疫抑制性細胞の誘導といった抗腫瘍免疫抑制的な環境形成の阻止にも有効で、既存治療の長期効果維持にもつながり、最終的には癌根治までが期待できるという点で様々な治療との相乗効果が期待できる。
