Outline of Research at the Start

高速な三次元生体組織イメージングが可能な光コヒーレンストモグラフィー (OCT) には横分解能が低いという問題がある。これを解決可能なデジタル収差補正には、通常の OCT よりさらに高速な撮影速度が求められる。
本研究では、生体の不随意運動に耐性のあるデジタル収差補正方法を開発して、高分解能 OCT 三次元イメージングを実証することを目的とし、
① 1次元走査分の OCT 信号のみでデジタル収差補正を行うアプローチ方法を提案して検証、
② 生体の OCT イメージングに対する提案手法の実証と問題点の検証
③ 提案方法におけるデジタル収差補正の最適化
を行う。

Outline of Annual Research Achievements

前年度までで達成された、1次元の収差補正と複数方向の走査データの組わせによる分解能向上に続き、OCT での高次収差の影響を表すモデル化、補正方法の検討、実際の in vivo ヒト網膜画像での実証と手法の検討を行った。
前年度で判明した、高次収差が複数ある場合の画質最適化の収束に関する問題に対し、高次収差がある場合の OCT 画像形成の理論モデルを精緻化して補正方法の検討を進めた。高次収差がある場合の OCT 画像生成理論を検討して、収差補正フィルターのモデルを検討し、その最適化パラメータの幾つかを物理量の予測に基づいて事前設定した。これにより、最適化に制約を加えることで、ある程度収束することが確認された。
これらの結果は、in vivo ヒト網膜のデータで確認された。ラスター走査パターンと、以前に開発した位相補正方法[K. Oikawa et al, Biomed. Opt. Express 11, 5886-5902 (2020).] によって、ある程度の狭い画角で収差補正した網膜の微細構造画像を取得できることが確認された。

2: Research has progressed on the whole more than it was originally planned.

Reason

今年度の予定であった、in vivo 生体での高次収差も補正するアプローチの動作検証と問題点の洗い出し・解決案の検討と実装が行われた。
これらにより、in vivo 生体での三次元 OCT 画像の高次収差補正が、狭い視野角という制限付きではあるが、達成された。
これらのことから、高次収差を含む場合の in vivo 生体高分解能 OCT イメージングと Lissajous 走査によるモーション補正・広画角化への事前準備がスムーズに進行しているため、本課題は当初の計画通りに順調に進展していると考えられる。

Strategy for Future Research Activity

モーション補正による安定した画像所得と、広画角化を目指し、Lissajous 走査と組合せた場合の高次収差補正の問題の洗い出しと、その解決方法の模索を進めていく。
また、より高分解能な画像取得を行うため、さらに大きな高次収差がある場合の補正方法の検討も合わせて進めていく。
