Outline of Research at the Start

Outline of Final Research Achievements
In this study, we aimed to develop artificial protein shells inspired by bacterial microcompartments (BMCs), which can autonomously and selectively encapsulate target proteins. We employed a combined strategy involving structural prediction, directed evolution co-expression screening, and electron microscopy analysis. Starting from the lumazine synthase (AaLS) derived from hyperthermophilic bacteria and the BMC shell proteins PduU and EutS from intestinal bacteria, we successfully created various functional protein shells with distinct sizes and properties. These results demonstrate the feasibility of using evolved BMC proteins for the rational design of artificial protein compartments, providing a promising platform for applications such as drug delivery systems.

Academic Significance and Societal Importance of the Research Achievements
