Outline of Research at the Start

本研究では、マウスES細胞と2CL細胞を用いて、全能性を規定する核内ゲノム3次元構造を明らかにし、多能性転換におけるMERVLの機能解明を目指す。まず、Hi-C法、LAD-seq、NAD-seqなどの手法によりES細胞と2CL細胞の核内ゲノム構造を比較する。また、アンチセンスオリゴによるノックダウンによりMERVLの機能阻害を行い、ゲノム構造への影響を解析する。

Outline of Final Research Achievements

This study investigated the nuclear function of MERVL, a retrotransposon transiently expressed in totipotent cells. Using ChIRP-seq in totipotent blastomere-like cells (TBLCs), we found that MERVL RNA is retained near its genomic loci, suggesting its nuclear localization after transcription. Furthermore, ChIRP-MS analysis identified several proteins interacting with MERVL RNA, most of which were spliceosome-related factors. Functional assays revealed that knockdown of hnRNP U or snRNP70 in ES cells upregulated 2-cell-specific genes such as MERVL and Zscan4c, and increased the number of MERVL-positive 2CL cells, without affecting pluripotency markers like Pou5f1. These findings suggest that MERVL RNA may interact with nuclear structural components, including the spliceosome and nuclear matrix, to modulate chromatin architecture and influence cell fate transitions during early development.

Academic Significance and Societal Importance of the Research Achievements

レトロトランスポゾンは、従来はゲノム不安定性の原因として抑制の対象とされてきたが、本研究はその転写産物であるRNAが核内において構造的・機能的役割を担い、幹細胞の全能性制御に寄与するという新たな概念を提起した。MERVL RNAがスプライシング因子や核マトリクスと相互作用し、クロマチン構造や2C遺伝子の発現制御に関与することは、非コードRNAによる細胞運命決定機構の理解を大きく前進させるものであり、再生医療や生殖工学などへの応用基盤としての意義も有する。
