Outline of Research at the Start

Outline of Final Research Achievements
Ferroptosis, an iron-dependent form of cell death, has been vigorously studied due to its relevance to cancer, neurodegenerative diseases, and ischemic conditions. In this study, we established a novel system that induces ferroptosis through iron supplementation in cells and used this system to screen for regulatory factors. Through this screening, we identified PRDX6 as a novel selenoprotein synthesis factor. In PRDX6-deficient cells, the expression of selenoprotein GPX4 -a master suppressor of ferroptosis- was reduced, leading to cell death upon iron treatment. Furthermore, we discovered that PRDX6 binds to selenium, the essential element for selenoprotein, revealing that PRDX6 functions as a previously unidentified intracellular selenium carrier.

Academic Significance and Societal Importance of the Research Achievements
