Outline of Research at the Start

近年加齢に伴う慢性腎不全患者の増加が大きな問題となっている。近年の慢性腎機能不全患者の増加には肥満、特にメタボリックシンドロームの増加が関与していると指摘されている。今回、我々はこのような病態で腎臓病理を生じるマウスモデルを確立したので、このモデルを使って腎機能低下のメカニズム、治療法の確立を目的に研究をする。本研究者はLKB1(STK11)という酵素の機能低下異常が主な原因であると仮説を持っている。今回の研究の目的は、腎臓病変の解析、 将来の遺伝子治療を念頭にヒトですでに広く使われているアデノ随伴ウィルス(AAV)を使いLKB1をターゲットに効果の検討をすることである

Outline of Final Research Achievements

1. The first year, I started feeding the mice with high fat and high sucrose diet to adipocyte specific MAPK1 KO mice (KO) and wt control mice (WT). KO mice increased body weight but its increase plateaued about 45 g in 3 months and 6 months. I and a PhD student (Dr. Ishinoda) examined the change in the kidney macroscopically and microscopically. The kidney side was not significantly different between KO and WT despite more increase in body weight. Glomerular size was also similar in size. And at this period, a lack of apparent morphological change indicated this model may not be kidney failure model observed in aging and obesity. 2. Over 14 month treatment, all the KO mice survived (5 mice) showed the tumor in the liver and shrinkage of the kidney, while WT mice showed none of these changes. These unexpected results further confirmed this KO mice model showed similar pathology of NAFLD in the liver and possibly in the kidney. These results were compiled for publication.

Academic Significance and Societal Importance of the Research Achievements

本研究では、様々な臓器に異常なほど高い脂肪蓄積を引き起こす独自のマウスモデルを用いて、肥満に関連する腎機能障害を明らかにしました。長期投与により肝腫瘍と腎萎縮が発現しました。このマウスモデルは、メタボリックシンドローム関連疾患の研究に有用でした。
