Outline of Research at the Start

ヒト脳神経系における重度損傷や難治性疾患などに対して、神経組織活動を人工的刺激により駆動・調節しようとする“神経補綴技術”は、近い将来、多様な神経機能不全を補償する一つの選択肢となる可能性がある。但し、その技術向上と安全性の確立には、対象となる神経組織の生理学的特性にも親和する、神経駆動刺激の新たな設計の方法論が求められる。この課題に対し本研究では、神経模倣工学的発想に基づく刺激生成器を開発し、これを用いた脳神経刺激の生理学的実験解析を通じて、「感覚器から脳へと入力される神経スパイクの時間系列を、脳組織へ与える人工的な刺激パルスの時間系列の設計に用いることが、如何なる合理性を持つか」を問う。

Outline of Final Research Achievements

To establish brain-based neural prostheses which compensate for dysfunctions of nervous systems in humans, it is essential to develop a methodology for designing artificial stimuli that effectively drive target neural circuits. Addressing the question, "To what extent is it rational to use the time series of neural spikes transmitted from sensory organs to the brain as a basis for designing stimulus pulse sequences applied to brain tissue?", we developed a system for novel retinal neuromorphic spike-train stimulation and conducted experimental analyses using millisecond-resolution imaging of cortical neuronal responses to the stimulation. The results suggested that, for instance, 1) the novel stimuli can drive neural activities while suppressing progressive inactivation of the neural circuits, thereby preserving their dynamic responsiveness, and 2) the elicited neural responses contain the information of temporal patterns of the light stimuli that induced the retinal spike trains.

Academic Significance and Societal Importance of the Research Achievements

本研究成果は、独自発想の神経刺激を実現する専用システムの新規開発に加えて、その新規刺激の有用性を実際の脳試料を用いた生理学実験を通じて実証したものである。いずれも、脳刺激型の視覚補綴研究においては、我々の知る限り国内外を問わず前例は無い。この神経模倣工学に基づく神経駆動法は、外界から網膜に与えられる光刺激の時間パターン情報を、脳皮質の神経活動へと効果的に反映させられる可能性を持ち、過去の臨床研究で用いられた固定周波数のパルス列刺激による光覚の誘発法に比べ、生理学的・情報論的な合理性を合わせ持つと考える。またこのことは、視覚補綴のみならず、他の神経機能補綴にも極めて重要な視点を与えると期待する。
