Outline of Research at the Start

生体分子機能の発現契機や作動原理は、分子全体の構造変化(アミド結合)と構成分子であるアミノ酸の局所的な分子振動変化の両方に支配される。それらの化学構造変化は分子の赤外吸収とラマン散乱によってそれぞれ観察できる。本研究では、1分子感度を有する探針増強型のラマン分光法と赤外吸収分光を共立する新奇なナノスケール振動分光法を駆使し、生体分子の機能と化学構造ダイナミクスの相関関係を明らかにすることを目的とする。

Outline of Final Research Achievements

In this study, we aim to elucidate the correlation between the function and chemical structural dynamics of biomolecules by utilizing a novel nanoscale vibrational spectroscopy technique that combines tip-enhanced Raman spectroscopy (TERS) with single-molecule sensitivity and infrared (IR) absorption spectroscopy.
We developed a multimodal optical microscopy system capable of simultaneously measuring spontaneous Raman scattering and mid-infrared absorption-induced photothermal effects. This system enabled concurrent acquisition of Raman and IR spectra from organic and polymer molecules.
As an application to single-molecule analysis, we performed high-sensitivity Raman spectroscopy of individual protein molecules and demonstrated structural analysis at the single-molecule level. We successfully identified the major secondary structural components of proteins, thereby obtaining structural information of proteins immobilized on a substrate.

Academic Significance and Societal Importance of the Research Achievements

生体分子機能に関連性が深い化学構造ダイナミクスを解析できる本技術は、タンパク質やペプチドが有する様々な生体機能を明らかにできる。多数の分子の信号を平均的に解析する従来の顕微分光では隠されてしまう1分子特有の挙動や不均一性も、分子を1つ1つ観測することで明らかになると期待される。加えて、グラフェンなどのナノカーボン材料の分析を得意とするラマン分光法とペプチドの2次構造やポリマー観察を得意とする赤外分光法を共立する本技術は、カーボン材料を混合したポリマー材料の化学構造評価にも有効であるため、それらを利用したデバイスの性能評価、新規開発に大きく貢献することが期待される。
