| Outline of Research at the Start |
オートファジーは細胞質成分をランダムに分解していると考えられてきたが、特定のタンパク質は集合状態の違いに応じて分解効率が変化することがわかってきている。本研究ではどのような状態の細胞成分がオートファジーによる分解を受けにくいかを調べる。質量分析を用いたオートファジー分解基質のプロテオーム解析により他の基質よりも分解されない基質群を選別し、分解されにくいタンパク質の共通性を調べることで、オートファジー分解を受けにくい細胞成分の状態を明らかにする。

| Outline of Final Research Achievements |
In this study, we identified glycogen-binding proteins as cargo that are poorly transported by autophagy. These proteins exhibited impaired transport due to interactions between bound glycogen molecules, causing them to behave as if they were larger than their actual molecular size. Similar characteristics were observed in nuclear and cytoskeletal proteins, suggesting that the "apparent size" within the cell, including intermolecular interactions, is a critical factor in determining the susceptibility of cargo to autophagic degradation.

本研究は、オートファジーによる分解において「対象となる構造の大きさ」が重要な制限因子であることを明らかにした。これまで注目されてこなかった分子間相互作用による見かけの大きさが輸送効率に影響することを示し、オートファジーの選択性の理解を深化させた。将来的には、異常タンパク質や細胞内構造の分解を促進する治療戦略において、標的の大きさや凝集状態を操作する新たなアプローチの基盤となる。
