Outline of Research at the Start

人工酸素運搬体(人工赤血球)は、輸血代替物や移植臓器保存液用途に加え、将来、3次元組織工学・再生医療の細胞灌流培養プロセスにも必要不可欠であるが、未だ実用化されていない。本研究では、ヒト赤血球のサイズ・形状や力学特性を模倣し、さらに超えることで、今までのPFC(パーフロオロカーボン)酸素運搬体と全く設計思想が異なる、ヒト赤血球様のサイズと形状を持つコアシェル構造を持った第4世代PFC酸素運搬体「cDFCs (concave-shaped Deformable PFC-based Oxygen Carriers)」の開発を試みる。

Outline of Final Research Achievements

The development of concave-shaped Deformable PFC-based Oxygen Carriers (cDFCs), which mimic the size, shape, and mechanical properties of human red blood cells, was promoted. cDFCs are core-shell particles with a red blood cell-like size and morphology, where the PFC core possesses the highest oxygen solubility among various solvents, enabling efficient oxygen absorption and release. Several oxygen carriers were successfully developed with different shell polymers, core solvents, and polymer coatings, featuring highly oxygen-permeable shells and reduced macrophage uptake. Furthermore, non-invasive in vivo dynamic evaluation was achieved using 19F MRI.

Academic Significance and Societal Importance of the Research Achievements

人工酸素運搬体は、将来、3次元組織工学・再生医療の細胞灌流培養プロセスなどに必要不可欠である。特にPFC型酸素運搬体は、酸素運搬能でヘモグロビン型に劣るものの、高い保存安定性や生体適合性から、ヘモグロビン型と並んで実用化への期待が大きい。本研究の酸素運搬体は、スケーラブルなプロセスで製造可能で、界面活性剤を用いていないために、界面活性剤起因の毒性懸念もなく、またコアのPFCのリークの危険性も小さい。さらに本研究では、in vitroの細胞培養プロセスへの応用や、in vivoにおける循環性能実証研究を実施し、実用化に向けて大きな技術的進展を得ることに成功した。
