Outline of Research at the Start

農薬中毒において、動物実験から推定された健康影響と実際の臨床現場で観察される中毒症状とにはしばしば大きなギャップがある。この原因として、従来型の毒性試験法では検出感度の問題から、その毒性を評価することが困難である事が原因の場合がある。本研究で提案するMITAシステムは、中枢神経の活動や代謝系の観察を、最先端のイメージング技術を用いることにより可視化する。それにより、連続的かつ高感度で、これまで見落とされていたAdverse Effectsを検出する。本システムにより、これまで困難であった連続観察や、最も感受性が高いと考えられる生きた胎児に対する薬剤影響の直接的な観察ができる。

Outline of Final Research Achievements

This study aimed to develop novel, highly sensitive, and non-invasive methods for evaluating the neurotoxicity and hepatotoxicity of chemical substances, with the goal of establishing the Multi Imaging Toxicity Assay (MITA) system. Using micro-computed tomography (micro-CT), we demonstrated that quantitative assessment of liver volume in rats is feasible and confirmed a correlation between increased liver weight/volume and marked induction of the CAR target gene Cyp2b1. Furthermore, lipidomics analysis of the cerebral cortex in mice exposed to the neonicotinoid pesticide acetamiprid revealed significant alterations in 15 lipid species, including fatty acids and FAHFAs. ROC analysis identified these as candidate biomarkers for neurotoxicity with AUC values greater than 0.9. These findings enabled the construction of a system capable of detecting early molecular signs of toxicity.

Academic Significance and Societal Importance of the Research Achievements

本研究は、micro-CTやリピドミクスといった先進的分析技術を毒性評価に導入し、薬剤による肝肥大や神経毒性に伴う分子レベルの変化を高精度かつ非侵襲的に検出可能とした点で、従来の毒性試験法を補完・革新する新たな枠組みを提示した。特に、FAHFAや飽和脂肪酸などの脂質の変動を指標とするバイオマーカー評価は、化学物質による影響の早期検出を可能にし、毒性発現機序の解明にも貢献する。また、動物を生かしたまま連続的に観察可能な手法は、3R原則を実現しうる有望な代替手段であり、将来的には国際的な化学物質リスク評価法への応用や標準化も視野に入る。これらの点から、本研究は学術的にも社会的にも高い意義を持つ。
