Outline of Research at the Start

腸内フローラは体内から24時間365日何10年間に及び人体に作用し続けるにも関わらず、その研究の多くは、一過的な影響を計るものがほとんどである。そこで、申請者らは、長期に及ぶ遺伝子発現制御に関わるエピゲノム修飾に着眼して、2型糖尿病が招く血管障害に起因する合併症の誘発あるいは抑制に関わる腸内フローラの因子探索を行う。本研究で明らかにされる、腸内フローラ成分によるエピジェネティックな遺伝子スイッチのオン・オフ情報は、長期にわたる高血糖状態に起因する血管障害に、腸内フローラが如何に関連しているかについて初めて明らかにするものである。

Outline of Final Research Achievements

This study aims to find gut microbiotic factor involved in the control of progression of type 2 diabetic (T2D) complications. First, gut microbiota was compared between T2D patient with and without lowered kidney function. As a result, Veillonellaceae and Prevotella were more abundant in the group of T2D patients without lowered kidney function. Veillonella is known to produce nitrite , which protects kidney function. Second, we investigated a series of epigenetic activity of major intestinal bile acids. As a result, a major secondary bile acid, lithocholic acid, shows a weak SIRT1 inhibitory activity and SIRT2 promoting activity, suggesting the possibility that lithocholic acid promotes progression of T2D complications through the epigenetic activity.

Academic Significance and Societal Importance of the Research Achievements

生活習慣病として増加の一途をたどる2型糖尿病であるが、その怖さは、自覚症状のないままに血管障害が進行し、神経障害、網膜症、腎症、動脈硬化など患者 のQOLを脅かす合併症に進展していくことである。本研究は、食と健康のインターフェースとして、人体に日々大量の代謝物を送り込む腸内細菌叢のエピジェノミックな作用に着目して研究を展開し、腸内細菌叢の主要代謝物であるリトコール酸は弱いながらも生体の代謝恒常性や炎症制御に働くサーチュイン活性に作用することを見出した。また細菌叢の因子として亜硝酸生産菌の関与の可能性も示唆した。さらに研究を進展させる意義ある発見であると考えられる。
