Outline of Research at the Start

初期胚はMHCの異なるホストの子宮に移植しても拒絶されない。これまで精巣の免疫寛容は精巣の体細胞が原因であると考えられて来た。しかし申請者らのCldn11欠損マウスへの移植結果や培養精子幹細胞の移植結果は、生殖細胞に免疫抑制のメカニズムがある可能性を示唆する。もし本研究にて精巣の免疫寛容のメカニズムが解明され、精子幹細胞のアロ移植による子孫作成が可能となれば発生工学の手法として新たな可能性を切り拓くものとなると共に、精巣の免疫寛容メカニズムの解明に新たな展開をもたらすものである。

Outline of Final Research Achievements

We successfully generated offspring by transplanting mouse germline stem cells (GS cells) into allogeneic hosts that are immunologically distinct in terms of major histocompatibility antigens. Specifically, GS cells established from testicular cells of DBA/2 mice were transplanted into either C57BL/6 or WBB6F1-W/W<sup>v</sup> mice as hosts. Remarkably, the transplanted GS cells were not rejected by the host immune system and were able to reinitiate spermatogenesis for more than six months. Mating these recipient males with wild-type females resulted in the successful production of offspring derived from the donor GS cells.

Academic Significance and Societal Importance of the Research Achievements

これまでメスにおいては胎児はアロ個体の中で発生して生まれてくることが知られていた。今回の我々の成果は精巣における免疫寛容を利用することでオスにおいても幹細胞から機能を持つ精子まで成熟させることができることを示すものである。ヒトにおいては倫理的な制約があるために成果を応用することができないが、実験動物においては精巣免疫の分子機構の解明に役立つ成果である。また免疫学的な解析が進んでいない動物種においての新規生殖工学技術の開発に寄与するものである。
