In the fiscal year 2023, significant progress was made in the research. The application direction of CRISPR imaging technology using labeled artificial chromosomes was determined, and a series of application explorations were conducted. Firstly, target sites suitable for CRISPR imaging were designed for Mouse artificial chromosome through genomic analysis. Through cell experiments, highly efficient labeling sites were screened, resulting in almost 100% labeling efficiency of artificial chromosomes in CHO and HT1080 cells. Secondly, various conditions were explored to optimize the labeling specificity of artificial chromosomes, significantly reducing the limit of detection. This laid the foundation for the application of this technology in the detection of rare chromosomes. Finally, a compatible high-throughput detection platform was established to detect artificial chromosome signals in live cells on a large scale. Currently, the detection throughput has reached 500 cells per second.In 日本分子生物学会2023, we have released some results by poster. The overall research is nearing completion, in the stage of organizing primary data and writing papers, aiming for the publication in influential international academic journals.
