Outline of Research at the Start

骨再生を担う骨芽細胞は骨髄や骨膜に存在する間葉系幹細胞(MSC)から分化する.人体には発生起源や形成過程が異なる種々の骨が存在し,四川大学Yuan教授らとともに,骨種や位置によるMSCの不均一性,ならびに加齢によるMSC細胞群の変化を明らかにする.また,遺伝子発現に基づき細分類されたMSC群と骨再生能とを紐付けし,MSCを再定義する.さらに,MSCの細胞挙動や運命決定におけるエネルギーセンサーの役割を解明し,新たな顎骨再生医療や骨粗しょう症治療の樹立へ向けた分子基盤形成を行う.

Outline of Annual Research Achievements

現在,間葉系幹細胞はと呼ばれているものは,実は単一な細胞ではなく多様性に富むヘテロな細胞集団であるため,コンセンサスの得られたMSCのマーカー遺伝子は定まっていない.間葉系幹細胞は骨や軟骨の間葉系組織再生のソースとして期待されているだけでなく,炎症の調整や拒絶反応の抑制に用いられることもある.よって,間葉系幹細胞を臨床応用する際の安全性の維持,品質管理の観点からも間葉系幹細胞がしっかりと定義される必要がある.
人体を構成する骨は,発生起源や形成過程が異なる206個の骨からなるが,異なる骨種間に存在するMSCの特性はわかっていない.そのため,例えば骨髄の脂肪化の起こりやすさは,骨の種類や場所によって異なることが経験的に知られているが,現時点ではこれを明確に説明することはできない.糖質を細胞に取り込み,産生されるエネルギーや代謝産物を細胞構成成分として利用することはすべての細胞に共通する根本的な生命現象である.しかし近年,ミトコンドリアにおける好気呼吸の代謝産物がDNAのメチル化を介して,破骨細胞分化を制御することがわかってきた.これらは単に破骨細胞のみならず,普遍的に細胞の運命決定や機能変化には糖質代謝状態,エネルギー代謝状態が直接的に関与する可能性を示唆する.本年度,マウス骨髄細胞をM-CSFとRANKL添加で刺激したところ,Gタンパク質共益型受容体の1つ,Tas1r3が破骨細胞分化とともにその発現量が上昇することを突き止めた.また脂肪細胞分化を強力に制御するCanonical Wntシグナルを強力に抑制する全く新規のペプチドを同定した.さらに脂肪組織代謝に関連するBMP3bが骨芽細胞系細胞にも発現し,骨芽細胞分化を制御している可能性を見出した.
