Research Abstract

エンドウヒゲナガアブラムシの無性生殖系統の起源に関してゲノムレベルの遺伝学研究を本年度も継続した。韓国で採集した個体について、核のelongation factor 1αの対立遺伝子をクローニングし、その配列を解析に加えた。解析の結果、交雑起源の無性生殖クローンが持つ２つの対立遺伝子のうち、一方の対立遺伝子と韓国の対立遺伝子が非常に近縁であることが明らかとなり、交雑個体の起源には大陸の集団が関与していることが示唆された。このことは、日本で繁殖している無性生殖クローンの起源はアジア大陸にあり、その起源がたいへんに古いことが示唆された。
エンドウヒゲナガアブラムシの人為的交雑個体が産むオスについて詳細に調べたところ、いずれの掛け合わせでもオス生産時のX染色体の分離比がランダムであること、オスの翅形成には常染色体上の遺伝子がかかわっていること、さらに、X染色体がオス胚子の発育に関与していることが明らかになった。両方の戻し交雑クローン合計12系統について有性世代誘導実験を行った。その結果、いずれの系統も有性メスを産出し、オスについては全く産出できないものから正常なオスが 産まれるものまで、クローンによって異なった。卵子形成時には遺伝子の組み換えが起こるため、ハイブリッドの作る卵の染色体では組換えが起こっていると考えられるが、組換え自体は有性メス産生には影響がないと言える。
シカハジラミの無性生殖の起源に関しては、新たに核遺伝子の７つの領域のプライマーを設計し、ミトコンドリアでの系統群間（Clade IaとClade IbとClade II）に変異があるかを調べた。しかし、2つの系統（Clade IとClade II）間の交雑の証拠は得られなかった。よって、シカハジラミの無性生殖では、交雑起源説は否定された。
