Outline of Research at the Start

TRPC3は活性酸素生成酵素(NADPH oxidase 2)とタンパク質複合体を形成することで心筋や骨格筋細胞の萎縮を誘導することを見出している。本研究では、TRPC3-Nox2複合体が病態時得意的に形成される分子機構を明らかにする。また、その結合を特異的に阻害する新たな化合物を同定し、筋委縮性疾患モデルマウスを用いてその有効性を検証する。
TRPC6チャネルについては、カチオン以外にFe2+やZn2+といった生命金属イオンも透過させることが知られている。そこで、TRPC6チャネルによる金属イオン流入が心臓の機能に及ぼす影響を明らかにし、これを標的とする新たな医療戦略を構築する。

Outline of Final Research Achievements

TRP channels are voltage-independent, cation-permeable channels that continue to attract attention as novel targets for next-generation ion channel-based drug discovery. We aimed to elucidate the isoform-specific physiological functions of receptor-operated TRPC channels and to establish a drug discovery strategy targeting isoform-specific TRPC protein. We demonstrated: (1) the TRPC6 channel exhibits permeability to essential metal ions such as zinc, which leads to improvement of chronic heart failure, inflammatory bowel disease, and hypertension; (2) TRPC3 protein forms a complex with the reactive oxygen species-producing enzyme Nox2 in a channel activity-independent manner, contributing to the progression of muscular dystrophy and the cardiac severity after COVID-19. Based on these findings, we propose a novel strategy for isoform-selective drug discovery targeting TRPC channels.

Academic Significance and Societal Importance of the Research Achievements

これまでTRPC3チャネルとTRPC6チャネルの両方が様々心血管疾患に対する悪玉因子と理解され、ひとまとめに阻害する創薬戦略だけが議論されてきた。我々はTRPC3タンパク質のチャネル機能に依存しない活性酸素/酸化ストレスへの相互作用の機序を解明し、TRPC3-Nox2複合体形成を標的とする新たな創薬戦略を提案した。一方、TRPC6チャネルの生命金属イオン透過性に着目し、TRPC6アイソフォーム選択的な活性化が心筋のZn2+動態を高め、心不全や炎症性腸疾患、さらには線維化疾患の改善に大きく貢献しうる可能性を動物レベルで実証し、TRPチャネル創薬成功の新たな戦略を提案した。
