Outline of Research at the Start

意思決定はヒトや動物にとって生存に必須な脳機能である。適切な意思決定のためには、良い選択肢を選ぶだけでなく、ときに悪い選択肢を回避する戦略をとることが重要になる。本研究では、戦略的な意思決定の神経機構を回路特異的な活動操作とイメージングによって明らかにする。さらに、精神疾患モデルマウスにおける戦略的な意思決定行動の異常とその神経回路病態を解明する。

Outline of Annual Research Achievements

本研究では、戦略的な意思決定の神経機構と精神疾患病態を明らかにすることを目的としている。昨年度までに戦略的な意思決定を必要とするマウス行動課題を開発し、意思決定行動中の側坐核のD1経路とD2経路のそれぞれに特異的な神経活動の制御とイメージングにより、D1経路とD2経路が異なる役割を担っていることを示した。そこで行った側坐核の一細胞レベルでの神経活動イメージングでは、D1細胞およびD2細胞のそれぞれにさらなる下位分類があることが想定された。そこで、本年度は側坐核の出力回路に着目して、下位経路の役割を調べた。側坐核D1経路は黒質網様部と腹側淡蒼球に出力している。そこで、側坐核D1細胞特異的にチャネルロドプシンあるいはアーキロドプシンを発現させ、黒質網様部あるいは腹側淡蒼球に光ファイバー経由で光照射によるターミナル刺激を行うことで、側坐核D1細胞-黒質網様部経路と側坐核D1細胞-腹側淡蒼球経路の二つの下位経路を分けて、活動制御を行うことが可能となった。側坐核D1細胞-黒質網様部経路と側坐核D1細胞-腹側淡蒼球経路のそれぞれの活性化はともにオープンフィールドでの活動亢進とリアルタイム場所嗜好試験およびタッチスクリーン報酬行動課題での快情動行動増加を示した。これらの結果は側坐核D1細胞-黒質網様部経路と側坐核D1細胞-腹側淡蒼球経路はともに運動と情動の制御を行っていることを示している。側坐核D1細胞-黒質網様部経路と側坐核D1細胞-腹側淡蒼球経路のそれぞれの活動抑制はともにこれらの行動に影響を与えなかった。

Strategy for Future Research Activity

側坐核経路の下位経路をそれぞれ分けて活動制御を行い、戦略的な意思決定を必要とするマウス行動課題における役割を解析する。戦略的な意思決定行動において活動した神経細胞をラベルし、その神経細胞への入力と出力を解剖学的に明らかにする。精神神経疾患モデルマウスを用いて、戦略的な意思決定行動における精神神経疾患病態とその神経機構を明らかにしていく。
