Outline of Research at the Start

1塩基置換編集ができるbase editor (BE)は、1塩基多型(SNP)変異で発症する多くの単一遺伝性疾患に対する根本的治療技術として期待されている。しかし、BEは活性の幅：activity window内に存在する標的SNP以外の塩基も置換するため、安全面での大きなリスクを抱えている。本申請では、安全性と治療効率を飛躍的に向上させるため、世界初となるactivity windowの精密制御による領域限定1塩基window編集技術を開発する。更に、未だ有効な治療法が無い遺伝性疾患モデルマウスの治療を通して、将来の臨床実用技術としての基盤を構築する。
