Outline of Research at the Start	われわれは、BIG3の細胞内局在がHER2陽性乳がんではトランス・ゴルジ網（TGN）に局在すること、さらに、抗HER2抗体薬トラスツズマブに耐性を獲得した難治性のHER2陽性乳がんではその局在が細胞膜へと変化することを見出した。この事実は、BIG3がTGNから細胞膜に移行することでトラスツズマブ耐性獲得に大きく関与することを意味し、細胞内局在が変化する分子メカニズムを解明することで、BIG3-PHB2複合体の制御を介したHER2陽性乳がん、特に詳細な分子機序が不明であるトラスツズマブ療法耐性に対する治療戦略を提案する。
