Outline of Research at the Start

本研究の目的は、Gタンパク質共役受容体(GPCR)の一種であり、睡眠・覚醒の制御に重要な役割を担っている「オレキシン2受容体」に対するGタンパク質バイアス型ペプチド作動薬の提案である。そのために、構造予測による分子モデリングや大型計算機による分子動力学(MD)シミュレーション、時系列データの時間情報を取り入れた動的解析手法などの計算化学・物理学を駆使して、リガンド選択性やGタンパク質・アレスチンシグナル選択性の動的機能機構を明らかにする。そして、得られた知見に基づきGタンパク質バイアス型ペプチド作動薬を設計・提案する。

Outline of Annual Research Achievements

研究期間を通じて、オレキシン2受容体 (OX2R) を中心に、アンタゴニストやアゴニストの複合体を含む不活性・活性状態および OX2R-Gタンパク質複合体の大規模なMDシミュレーションを実施し、動的解析手法である緩和モード解析によりTM3の特徴的な構造変化と中間状態であるNon-canonical stateを抽出し、定量評価した。解析結果をデータベースと統合して解析することにより、リガンド選択性とシグナルバイアスに寄与する残基を同定し、OX2R に対するアゴニスト設計に有用な知見を得た。さらに手法をκオピオイド受容体など他のクラス A GPCR に展開し、バイアスシグナリングのメカニズムを示唆した。成果として国際誌2報掲載(うち1報はJournal front coverに選出)、3報投稿中、国内外計13件の発表と3件の招待講演、3件の記事執筆を行い、CBI学会から口頭発表賞、分子シミュレーション学会から学生発表賞、International Union of Pure and Applied Physics (IUPAP)からTaylor & Francis Molecular Simulation Prize、GPCR研究会から松尾研究奨励賞を受賞した。Stanford University、SLAC National Accelerator Laboratory、Koc University との国際共同研究も推進し、Nature Communicationsへ国際共著論文の掲載などに結実した。
