Research Abstract |
To develop detection method for viable but non-culturable (VBNC) bacteria, we investigated molecular mechanism of VBNC state. For analysis of cell cycle of Salmonella, visualization method of DNA replication origin by FISH was established. While 1 or 2 foci of replication origin was detected in log phase cells, we failed to stain replication origin in VBNC cells. In VBNC state, physical state of chromosomal DNA might be different from that in log phase. Expression levels of FtsZ protein, which forms cell division ring, were not significantly different between stationary, log phase, and VBNC state. This indicates that VBNC state is not presumably induced by cell cycle arrest by reduction of FtsZ expression levels.
