Research Abstract |
To clarify molecular mechanism of C/EBPa transcriptional regulation in granulopoiesis, we tried to identify transcriptional cofactors of C/EBPa. As a result, we identified poly(ADP-ribose) polymerase-1 (PARP-1) as an interactant of C/EBPa. PARP-1 co-represses C/EBPa transcriptional activity and reduces gene expression of C/EBPa target gene, myeloperoxidase (MPO). Additionally, PARP-1 binds C/EBPa response elements region and colocalizes with C/EBPa there. It is supposed that PARP-1 inhibits C/EBPa binding to response element.
