Outline of Annual Research Achievements |
平成28年度は、オールインワンナノ治療デバイスとして、温度応答機能、MRIや近赤外外蛍光イメージングによる可視化機能に加えて標的病巣がん細胞へのターゲティング機能を持たせるために、CD20陽性Bリンパ腫に特異性をもつリツキシマブを結合した多重機能リポソームや、金ナノロッドーデンドリマー複合体を作製し、そのキャラクタリゼーションと細胞実験を行った。リポソームや金ナノロッドハイブリッドにリツキシマブを結合することで、CD20陽性細胞へのキャリアの取り込みが大幅に上昇し、加温下や近赤外レーザー照射下において封入した抗がん剤が放出されることで高い細胞成長阻害効果が見られた。また、金ナノロッドと非対称デンドリマーからなるオールインワンナノベクターの開発について、粒径40nm程度の金ナノロッドと温度応答性高分子の側鎖構造を修飾した非対称カチオニックデンドリマー(デンドロン脂質)を複合化させたナノハイブリッドを作製した。このナノハイブリッドは、プラスミドDNAと効率よく複合体を形成した。また近赤外レーザーを照射すると金ナノロッドが発熱し、デンドロン末端の温度応答性側鎖が疎水化し、キャリア表面の疎水性が増大することで、細胞により効率良く取り込まれ、高い遺伝子発現を導いた。このことから、光・熱応答型遺伝子ベクターとして機能することがわかった。さらに、オールインワンナノワクチンとして、pH応答性基を導入した多糖誘導体被覆リポソームに、カチオン性脂質を組み込むと、従来の脂質アジュバントであるMPLAよりも極めて効果的に樹状細胞を活性化し、CTLの誘導に重要なサイトカインIL-12が大量に産生された。また、MPLAを含有したpH応答性ポリマー・pH応答性デンドロン脂質修飾リボソームをがん抗原ペプチドのデリバリーシステムとして用いるとそれぞれCTL・ヘルパーT細胞の誘導効率が増大することも明らかにした。
