Outline of Final Research Achievements

Recently, the fungus Penicillium cyclopium was found to produce conidiogenol and conidiogenone as self conidiation inducing-diterpenoids. This finding indicates that a novel diterpene synthase would utilize for the biosynthesis of these diterpenoids. To study the conidiation in fungi, we have identified two novel diterpene synthases (PeDS and AnDS1) from P. cyclopium synonym and Aspergillus nidulans A4. Heterologous expressed in vivo enzyme assays revealed that PeDS catalyzes the GGDP synthesis and convert into novel 3-deoxyconidiogenol, and AnDS1 catalyzes the GGDP synthesis and into novel diterpene hydrocarbon, respectivery. The structures of two novel compounds were elucidated by spectral data. These compounds were revealed to have conidiation inducing activities in the submerged culture of P. cyclopium synonym and A. nidulans A4, respectivery.
