Outline of Final Research Achievements |
Ten plaques removed from carotid endarterectomy patients were classified into high- and low-calcified plaques on the basis of Agatston calcium score. Microarray analysis for miRNA profiles was performed followed by validation with miRNA quantitative real-time (qRT) PCR analysis. Genes which satisfied total gene signal (TGS) >50 and [Log2 ratio <-1 or >1 were selected. Among carefully selected 70 angiogenesis or calcification-related transcripts, reliably expressed miRNAs marking median and high context scores with miRSearch and FDR <0.1 by the two-group comparison were also extracted .In high-calcified carotid plaques, specific profile for miRNA may be shown and expression of hsa-miR-4530 and hsa-miR-133b had inverse correlation with calcium score in plaques, suggesting miRNA may play a modulating role in calcification in plaque and plaque stability.
