Research Abstract

Genomic clones that contain the <alpha> -globin of the marine fish yellowtail (seriola quinqueradiata) were isolated by cross-hybridization to <alpha> -globin cDNA of carp from the yellowtail genomic library. The <alpha> -globin gene of yellowtail with the length of 1.6 K bases was cloned. The endonuclease restriction map of <alpha> -globin gene of yellowtail was almost similar to <alpha> -globin gene of carp. we determined the complete nucleotide sequences of the <alpha> -globin gene of carp containing 2 introns and 3 exons. The length of the <alpha> -globin gene from the codon initiating translation(ATG) to the terminal codon(TAA) was 714 base pairs(bp). From the 5' leader sequences to the 3' untranslated regions, the first 96 bp nucleotide of the <alpha> -globin gene are encoded in a single exon, followed by intron 1, a second exon domain extending to codon 264, a second intron, and a third exon domain(134 pb) comprising the rest of the transcribed DNA. The first intron(176 bp) of <alpha> -globin is slightly larger than the second intron(106 bp). Similar sequences are found in the 3' untranslated region and the introns in correponding locations in most mammalian <alpha> -globin genes.
