| Research Abstract |
ラット肝から分離精製したグロボトリアオシルセラミド合成酵素(αーGal転移酵素)を中心に研究した.
(1)^3H-UDP-dialdehydeを合成し, α-Gal転移酵素を標識し, 酵素の大小2つのサブユニットのどちらかに標識されるかを検索した. また, 100%不活化時における化学量論的な結合量をみた.
(2)^3H-UPP-dialで標識した酵素をTPCK-トリプシンで消化し, 標識ペプチドを高速液体クロマトグラフィーおよびペーパークロマト, 高圧電気泳動で分画した.
(3)標識ペプチドのアミノ酸組成および一次構造を, シークエンサーで解析し, アミノペプチダーゼM, カルボキシペプチダーゼA, エンドリジルペプチダーゼなどで処理し, 標識アミノ酸を同定した.
