Research Abstract

The purpose of the present study is to examine the biological significance of myelin basic protein (MBP) immunoteactive substance in tumor cells. We examined at randow many tumors of carious histolobic types and degree of differentiation arising in various organs and tissues fixed in 10 to 20% formalin and embedded in paraffin from surgical and autopsy materials. The investigative procedures were immunohistochemical and immunoelectron microscopic mwthods by using polyclonal MBP antibody and indirect immunoperoxidase technique. The tumor specimens for immunoelectron mictroscopy were surgical materials taken at ehr time of frozen section diagnosis and were fdixed in 4% buddered parafgormaldehyde soultion. ALL tumors contained MBP immunoreactive cells, which varied considerably in numver form scase to cvase and from area to area even in the same case. The intensity and nature of staining were variable form cell to cell. Immunoelectron microscopically the MBP immunoreactive substance was observed in various organelles including membrane-attached ribodomes, free ribosomes, vesicles, and cell surface. Perinuclear space, discternae of rought endoplasmic reticulum, Goligi apparatus, and mitochondira were negative. Nuclei rarely contained immunoreactive substance. The present results suggest that tumoe cells generally produce MBP immunoreactive substance, regardless of histologic typing, Degree of differentiation, and source of tumors. The exact biochemical nature of MBP immunoreactive substance couls not be determined in this study. However, we wolud like ot postlate that the substance could well be a mitogenic polyperptride for tumoe cells, consifering that a bovine brain derived polypeptide having somewhat similar amino acid composition to MBP is known to be mitogenic for vatious types of cells and is called fibroblast growth factor (FGF).
