Outline of Annual Research Achievements

病原性微生物がホスト細胞に障害を与えるために産生する膜孔形成毒素は,可溶性の単量体として分泌されるが,ターゲット細胞に接すると膜上でpreporeと呼ばれる円状の会合体を形成した後,劇的な構造変化を起こして膜孔を形成する.黄色ブドウ球菌は,3種類の非常に類似した膜孔形成毒素(Hla,LukF,Hlg2)を分泌するが,Hlaはホモ7量体の膜孔を形成するのに対し,LukF,Hlg2は交互に会合したヘテロ8量体の膜孔を形成する.毒素同士がどのようにして特定のパートナーとのみ特異的に会合して膜孔を形成するのか,また,7量体・8量体という分子の会合数がどのようにして決定されるのかについては,明らかにできていない.
本研究では,構造解析から提唱された機構を基に変異体蛋白質を合理設計し,毒素の会合特性を改変することを通して,これらの毒素の自己組織化機構を解明することを目指した.立体構造情報をもとに,Hlg2とLukFの接触界面を移植したキメラ変異体を2種類作成した.また,コンピュータシミュレーションにより単量体として機能できるように,界面の残基の変異体を設計した.4種類の変異体を作成した.これらに加え,2成分性毒素の各成分に対し,ステム領域をHlaのもので置換した変異体と,さらにその周辺の重要残基に変異を加えたキメラ変異体を合計16種類作成した.これらのキメラ変異体の溶血活性を種々の条件下で評価した結果,LukFのステム領域はHlaのステム領域で置換可能であることがわかった.また,Hlaのステムで置換したHlg2の種々の変異体のCDスペクトルから,ステム領域の入れ替えにより,単量体の構造に変化が起きていることが示された.また,一連の実験の過程で, Hlg2,LukFのリフォールディングの手法が確立された.
