Outline of Annual Research Achievements

【本研究の目的】中分子化合物を医薬品や研究ツールとして活用する試みが近年盛んに行われている。本研究では、改変翻訳合成系を用いた大規模中分子ライブラリーの構築と試験管内選択を行い、任意の生物活性を有する中分子化合物をテーラーメイドに『創製』する新技術の確立を目標とする。具体的には、Targeted Covalent Inhibitor(TCI)として機能する環状ペプチド中分子の創製を目指す。
【研究実績の概要】TCIを開発する方法論として、本計画では、広い作用面を介して標的と相互作用できる中分子の試験管内分子進化法の活用と、共有結合形成能を自在に調節できる反応性官能基(tunable Reactive Group, tRG)を新たに開発することを提案してきた。具体的には、A. tRGの開発とカタログ化、B. RGを有する環状中分子ペプチドライブラリーの構築と試験管内選択システムの確立、C. tRGによる中分子TCIの反応性調節戦略の実証、という三つのマイルストーンを設定し、これらを順次達成することで、上述の最終目標に到達する計画である。
本年度の研究では、前年度に開発を完了したtRG含有アミノ酸を用いて、環状中分子ペプチドライブラリー を構築することに成功した。また、実際に試験管内選択実験により新規共有結合性ペプチド阻害剤の取得にも成功した。さらに、このペプチド阻害剤中のtRG構造を変えることで、共有結合形成能を調整できることも実証した。以上により、本研究が標榜していた共有結合を介して標的を阻害する高選択性中分子阻害剤の汎用創製手法を確立できたと言える。
