Outline of Annual Research Achievements

本研究課題では、耐性能力の異なる複数のクマムシのゲノム・トランスクリプトームデータを基に新規耐性遺伝子の探索を進めている。本年度は、乾燥耐性の非常に高いクマムシ種で常時高発現し、耐性誘導型のクマムシでは乾燥暴露によって発現が有意に誘導される新規タンパク質を同定し、RNAi 法を用いて同タンパク質のクマムシの乾燥耐性への寄与を検証した。同タンパク質は相同性検索においてクマムシ類の一部の配列とは明瞭な相同性を示すものの、クマムシ以外の生物に由来する配列には相同性が見出されず、クマムシ類に固有のタンパク質であることが判明した。耐性の高いクマムシの同タンパク質に対する抗体を作成し、細胞内局在を解析した。また、同種では RNAi による耐性への影響は観察されなかったが、耐性誘導型のクマムシを用いたRNAi実験では、乾燥・給水を特定の条件で行った際に乾眠後の回復率が有意に低下することが分かった。同タンパク質は乾燥からの生体分子の保護に寄与していることが考えられる。また、前年度までに同定していたクマシム固有のDNA防護タンパク質Dsupについて、より詳細な生化学的な解析を行ったほか、DNA やヌクレオソームと混合した状態で分子間力顕微鏡を用いた解析を行い、それぞれに対する結合様式を明らかにした。特にヌクレオソームに対してはDNA単体よりもより低濃度で顕著な形態変化を誘導することを見出し、Dsupがヒストンと協調してDNAに結合/高次構造を変化させることを明らかにした。
