Outline of Annual Research Achievements

A3G及びA3Fはレトロウィルスが合成するssDNA上のシトシンをウラシルに脱アミノ化することにより遺伝情報を破壊する。本研究では、A3G及びA3Fが一本鎖DNA中の標的シトシンを探索する際の分子機構の解明を目指して来た。これまでA3Gについて解析を進め成果を上げてきたが、今回はA3Fの解析を行い論文発表した[BBA, 2020]。まず、A3FのN214H変異体の活性が高いことを見出し [PCCP, 2018a]、これを使うことによりNMR解析が可能であることに思い至った。そして、これまで用いてきたuracil-DNA glycosylaseアッセイに加え、実時間NMR法も用いて脱アミノ化酵素活性を調べた。これにより、A3Fは濃度条件[A3F-CTD]≫[ssDNA]では長いssDNA中の標的シトシンを効率よく脱アミノ化するのに対し、[A3F-CTD]≪[ssDNA]条件では短いssDNA中の標的シトシンを効率よく脱アミノ化することが明らかとなった[BBA, 2020]。さらに本研究では、脱アミノ化速度とssDNA結合親和性を使って半定量解析することにより、A3Fのこの特徴的な活性について合理的に説明することにも成功した[BBA, 2020]。他方、機能性核酸の細胞内動的構造解析については、これまでにヒト培養細胞に導入した核酸のin-cell NMRに成功していた[PCCP, 2018b]。核酸のイミノ水素は塩基対を形成したときのみNMRシグナルとして観測される。したがって、イミノ水素と水の水素の交換速度を知ることにより、核酸構造の局所的な安定性を評価することができる。我々は、ヒト培養細胞に導入した核酸についてこの交換速度を測定するin-cell NMR法を開発し、核酸構造の安定性に関して議論することに成功し、第18回蛋白質科学会年会において口頭発表を行った。
