Outline of Annual Research Achievements

医薬品は化学物質であり、低分子であれば簡単な化学構造式で表現することができる。しかし、我々は化学構造式を見ただけで如何なる薬理作用を発揮する物質であるかを当てるほど十分な知識を持ってはいない。本研究では薬→作用点への親和性を予測する人工知能を開発した。学習に用いた化学構造と作用実測値はChEMBL25 (March 2019)から抽出し、ChemAxonを用いてSMILES形式データを作成した。作用点としては、医薬品作用点の構造分類に鑑みて、右表に示す数の実測値を有する20種類の標的分子を選択した。深層学習はDeepChemライブラリに含まれるグラフ畳み込みニューラルネットワーク(GCNN)を用い、実測値データを8:1:1に3分割して8を学習に、1をハイパーパラメタの最適化に、残った1を最適化後の予測値と実測値の相関性プロットに用いた。その結果、抗うつ薬標的であるセロトニントランスポータ、統合失調症治療標的であるドパミンD2受容体、無痛症の原因遺伝子で鎮痛薬標的とされる電位依存性Na+チャネルSCN9A (Nav1.7)など、20種類の作用点のうち17種類に対して決定係数0.6以上の予測モデルを構築できた。次にこの予測モデルの妥当性を生物学的に検証した。SERT阻害薬は何らかの抗うつ作用を有することから、ChEMBLに収録されたうちSERTに対する作用が調べられていない化合物を抽出し、構築した予測モデルを用いてSERT阻害活性を予測した。そのうち約10 nMのIC50を有すると予測された化合物1について、抗うつ作用スクリーニングに用いられる尾懸垂試験および非特異的な運動活性亢進を検知するためのオープンフィールド試験を行ったところ、化合物1は10 mg/kgの用量において自発運動活性に影響することなく、有意な抗うつ作用を引き起こした。
