Research Abstract

我々は病原性細菌が共通して持つ病原性発現機構(病原性発現コアシステム)の理解を目指し、黄色ブドウ球菌のカイコ感染モデルを用いて、細菌間で保存された新規の病原性遺伝子cvfA、cvfB、cvfCを同定した。本研究では、CvfBの結晶構造解析を行い、CvfBの分子機能を明らかすることを試みた。肺炎レンサ球菌のCvfBを1.4Åの解像度で結晶構造を明らかにした。CvfBは、3つのS1 RNA結合ドメインと1つのWinged-Helix(WH)ドメインからなるL字型の構造を有することが明らかとなった(2)。肺炎レンサ球菌と黄色ブドウ球菌のCvfBは、いずれもpoly(U)結合活性を有した。poly(U)との結合には、CvfBのC末端側のS1 RNA結合ドメインとWHドメインが必要であった。結晶構造からWHドメインの3つのリジンがRNAと相互作用することが予想されたこと、ならびにリジンに変異を導入したCvfB(K249A/K267A/K275A)がpoly(U)結合活性を低下したことから、WHドメインのリジン残基がRNAとの直接結合に必要であることが示唆された。さらに我々は、CvfB抗体による免疫沈降により、CvfBの相互作用因子として、30Sリボソームの構成タンパク質であるS9を同定した。CvfBとS9の結合には、CvfBのWHドメインが必要であった。WHドメイン欠損CvfBをコードする変異型cvfBは、黄色ブドウ球菌のcvfB遺伝子破壊株における溶血毒素の産生量低下を相補しなかった。したがって、CvfBはWHドメインを介してRNAならびにリボソームと相互作用し、溶血毒素産生に寄与すると考えられる。以上の結果は、CvfBが翻訳段階で病原性遺伝子の発現を調節するRNA結合タンパク質であることを示唆する。
