Research Abstract

我々は、小脳プルキンエ細胞に発現する1型代謝型グルタミン酸受容体mGluR1と別種のGタンパク質共役性受容体(B型GABA受容体GABAbRおよびA1アデノシン受容体A1R)の機能的連関の生理的意義と機序の解明を目指している。前年度の研究で示唆されたGABAbR活性化がmGluR1介在性の視機性動眼反射(OKR)順応(長時間動く視覚パターンに暴露することで、パターンに対する眼球の追随性=OKRゲインが向上する学習)を促進する可能性を、別の実験者チームに検証させ、上記観察が高い再現性を有することを明らかにした。トレーニング初日のOKRゲイン増加の時間的推移にはGABAbRアゴニストを小脳片葉に注入したマウス群と統御群との間に差異が認められなかったが、トレーニング2日目において注入群は初日に達成したゲインを保持していたのに対し、統御群は保持していなかった。同じ注入法により局所麻酔剤を投与するとトレーニング初日のOKRゲイン増加が消失したことから、小脳片葉のOKR順応に対する関与が確認された。また上記実験で用いた自主開発のOKR測定装置が極めて高い精度で学習成績を評価できていることが確認された。これらの結果は、GABAbRの活性化がOKRに関する長期記憶の固定プロセスを促進する可能性を示唆している。またin vivoの小脳片葉で誘発された小脳長期抑圧を、シナプス画分のグルタミン酸受容体GluR2サブユニットの減少として捉えるimmunoblot解析方法を確立し、in vivoにおけるGABAbR活性化の小脳長期抑圧に対する効果を精査中である。一方、上窪らと共同でNeuro2a細胞株に発現させたmGluR1とA1Rが機能的および物理的に相互作用することを示唆するデータを得た。この結果は、これら受容体の相互作用がニューロン種特異的な細胞内環境に依存しないことを示唆している。
