Research Abstract

1)SGS3はRNA-dependent RNA polymerase 6(RDR6)と協働してウイルス由来のRNAに対する小分子RNA、内在性のTAS遺伝子を前駆体とする小分子RNAの生合成に関わる。このSGS3とRDR6が相互作用すること、さらに新規な構造体SGS3/RDR6-bodyとなづけた構造に局在することが明らかとなった。mRNAの分解や貯蓄を担うProcessing-body(P-body)とは独立の構造であった。このことで、tasiRNA生成の過程で中間体が細胞内で複雑に、複数の会合体間を移動することが示唆された。今後一過的な発現ではなく、ナチュラルに近い状態での発現におけるSGS3/RDR6-bodyの存在、組織依存性、発生時期依存性を明らかとする必要性がでてきた。
2)免疫沈降法によるAGO1と結合する3種類の候補タンパク質の解析をおこないかたわら、東京大学大学院理学系研究科の上田貴志氏に種々の細胞内膜系のマーカーを発現するラインをいただき、詳細なAGO1あるいはP-bodyとの関係を解析した。
3)AGO1,RDR6タンパク質の発現の組織特異性、発生時期特異性を検討するために、各遺伝子のプロモーター(2-3kb)下流にGUS遺伝子をつなぎ、形質転換体を作成した。双方の発現はメリステム近辺で強いが、AGO1タンパク質は発芽後一週間ほど、RDR6はむしろそれに遅れて10日過ぎに発現が多くなることを見いだした。今後SGS3についても同様のプロモーター解析を行い、SGS3/RDR6-bodyの時空間的機能解析の重要性が明らかとなり、解析を継続している。
