Outline of Research at the Start

NMRは、X線やCryo電顕では困難な溶液条件下での構造解析を得意とし、細胞内の蛋白質を細胞が生きている状態のまま構造解析できる唯一の手法である。しかし、生細胞内での蛋白質の構造解析は再現性や検出感度に問題があり、ヒト細胞での成功例はない。そこで、本研究では独自技術で超高感度化に成功したin-cell NMR法を発展させ、ヒト生細胞内蛋白質の立体構造解析技術を開発する。また、応募者らが独自開発した液-液相分離状態における蛋白質の構造解析技術などと組み合わせ、生細胞内におけるセラミド輸送蛋白質CERTとEP局在足場蛋白質VAPからなるER-Golgi膜接触領域の立体構造解析を目指す。

Outline of Annual Research Achievements

NMRは、X線やCryo電顕では困難な溶液条件下での構造解析を得意とし、細胞内の蛋白質を細胞が生きている状態のまま構造解析できる唯一の手法である。本研究では、「ヒトの細胞内で働いている蛋白質の姿は我々の知る蛋白質構造と同じなのか?」という問いに答えるため、(1)ヒト生細胞内蛋白質の立体構造解析技術の開発、(2)LLPS蛋白質のヒト生細胞内立体構造解析技術の開発、(3)セラミド輸送蛋白質CERTとEP局在足場蛋白質VAPからなるER-Golgi膜接触領域のヒト生細胞内立体構造解析、の3つの課題に取り組んできた。各課題に関する本年度の研究実績は以下の通り。

課題(1):ヒト生細胞内の蛋白質の立体構造変化やリン酸代謝を分単位精度でリアルタイム検出する技術の開発に取り組み、蛋白質間相互作用や蛋白質薬剤相互作用など分子間相互作用についても解析を進め、細胞内小胞内の酸性化プロセスのリアルタイム検出に成功した。
課題(2):LLPS状態のVAP蛋白質の真空紫外円偏光二色性スペクトルを測定・解析し、応募者らが決定した溶液NMR構造や結晶構造と比較した。LLPS状態と溶液状態で立体構造に大きな違いはなかったものの、明確な構造の違いが検出された。
課題(3):Cyo-EMを用いてCERT蛋白質全長にフォーカスを絞って立体構造解析を進めた。クライオでは像が得られなかったが、ネガティブ染色像からCERT蛋白質全長が3量体を形成していることが示唆された。
