Research Abstract

遺伝的揺らぎ研究:これまで、肺癌検体を用いたmRNAマイクロアレー解析、細胞レベルにおける各種実験結果より、POLD4遺伝子の重要性を多角的に解明してきた。POLD4発現低下は細胞レベルで、肺癌発生と密接な関連を有するタバコなどによるDNA損傷修復活性の低下をもたらす。外的損傷がない状態でもG1/S期およびS期における細胞周期チェックポイント活性化を誘発し、染色体断裂頻度の上昇を伴う。平成25年度はこのチェックポイント活性化につながる機序の解明を行った。すなわち、POLD4発現低下はDNA複製・修復能低下により2本鎖切断を誘発し、ATMの活性化を引き起こす。その結果p27遺伝子産物の分解抑制によりチェックポイントの活性化が導かれることが明らかとなった。
細胞膜揺らぎ研究:ハイブリッドリポソーム処理により、種々のがん細胞においてアポトーシスが誘発される。我々は、その過程で本来細胞内に極めて微量にしか存在しないリン脂質が増加すること、また、そのリン脂質の下流代謝物のひとつである、スフィンゴ脂質組成が変化することを明らかにしてきた。さらに、これらスフィンゴ脂質組成変化の責任遺伝子を同定したところ、その責任遺伝子が癌で高発現していることが明らかとなった。この責任遺伝子の癌における機能を明らかにするため、ノックダウン、過剰発現を主体とした実験系を構築し、解析を行った結果、この遺伝子は癌細胞の転移に必要であることが明らかとなった。以上の結果はハイブリッドリポソームは、癌転移能亢進のため細胞が高発現している遺伝子産物の活性を利用して、抗癌剤としての作用を発揮するという機序を示している。
