Outline of Annual Research Achievements

血管が形成する神経幹細胞niche(vascular niche)への理解は非常に重要な研究課題である。しかしながらvascular niche の本質はいまだ明らかになっていない。我々はendothelial microparticles (EMP)が神経幹細胞のstemness維持に及ぼす作用を検証し、vascular niche の本質に迫ることを本研究の重点課題とした。
血管内皮細胞と神経幹細胞との共培養系を確立し、幹細胞マーカーの発現を解析した。血管内皮細胞と神経細胞を共培養すると、興味深いことに、神経幹細胞のstemness保持が促進され、神経分化が抑制される傾向がmicroarray解析、免疫細胞化学的解析の結果明らかとなった。血管内皮細胞が産生するendothelial microparticles (EMP)が神経幹細胞の分化をコントロールしている可能性が考えられたため、共培養系におけるmicroparticle画分のmiRNA array解析を行った結果、神経幹細胞の分化を抑制するmiRNA群の発現が観察された。以上の結果から、血管内皮細胞が形成する神経幹細胞nicheの一つとして、EMPに包含されるmiRNAが重要であることが示唆された。
