Research Abstract

本研究ではES細胞からの立体組織培養系と3次元イメージング技術を組み合わせ、眼杯形成についての分子・細胞・組織の各階層をまたいだ解析を行うことで、上皮組織の形態形成制御機構について新たな知見を得ることを目的とした。二光子顕微鏡を用いて眼杯形成過程における個々の細胞動態やアクチンや微小管などの細胞骨格動態を長期にわたってイメージング出来る実験観察系の構築を行った。その結果、領域特異的な細胞増殖スピードの違いや細胞形態変化が明らかになった。また同時に細胞内カルシウム動態を観察することで、眼杯形成過程における領域特異的な細胞形態変化と自発的な細胞内カルシウム動態との相関関係を明らかにした。こういった実測データを三次元上皮組織の力学的特性を検証するための数値シミュレーションモデルと組み合わせることで眼杯形成の基本原理についてより厳密に検討することが可能になった。本モデルにより自発的な揺らぎといった個々の細胞動態と組織変形の関係についての階層を超えた理解がさらに深まることが期待される。同時に、ヒトES細胞から自己組織化的にin vitroでヒトの眼杯組織および大脳組織を構築することにも成功し発表した。in vitroでのヒト網膜組織分化法の開発により、将来のヒト網膜組織を用いた、網膜色素変性症などの眼疾患の移植治療の実現化に大きく近づくことになると考えられる。また、in vitroでヒトES細胞から分化させた大脳皮質組織では、発生過程に見られるような層構造の形成が確認され、同時にヒト大脳皮質発生過程に特異的に存在する細胞(basal progenitor)も確認された。
