Research Abstract |
アクチン骨格はアクチン繊維と多様なアクチン結合蛋白質から構成される過渡的複合体として細胞内に存在しており、この複合体の形成と崩壊の時空間的制御は細胞の運動、形態形成、分裂などに必須である。コフィリンはアクチン脱重合因子として、アクチン過渡的複合体のダイナミクスを制御する重要な因子であり、その活性はLIMキナーゼによるリン酸化とSlingshotによる脱リン酸化により制御される。本研究では、細胞運動時のアクチンダイナミクスの制御機構の解明を目的として研究を行い、以下の成果を得た。 1、可逆的な光活性化蛍光蛋白質Dronpaを用いて、生細胞内の局所におけるG-アクチン濃度の時空間的変化を定量的に測定するsequential FDAP (s-FDAP)法を開発し、増殖因子刺激依存的な細胞内G-アクチン濃度変化を解析し、刺激前の細胞内G-アクチン濃度が、刺激後のアクチン重合度及び仮足伸長度を決定する重要なパラメーターであることを解明した。また、メカニカルストレスによるアクチン骨格の再編成において、G-アクチン濃度の変動が重要な役割を担っていることを明らかにした。 2、新たなBiFCプローブを開発し、LIMキナーゼ阻害剤を探索し、Damnacanthalなど2種の阻害化合物を同定した。Damnacanthalは、LIMキナーゼの阻害を介して、癌細胞の浸潤やマウス樹状細胞の遊走を阻害することを見出した。 3、Slingshotの結合蛋白質をプロテオーム解析により同定し、Slingshot-1S及び-1L アイソフォームに特異的に結合する蛋白質の存在を明らかにした。
