Outline of Annual Research Achievements

①両親媒性高分子の疎水部にROS消去能を有するニトロキシドラジカルを結合し、その水中での自己組織化による40nm程度のナノ粒子を作製した(RNP)。このRNPをナノ反応場とし、TEOSの可溶化とゾルゲル法によりシリカ含有レドックスナノ粒子siRNPを作製した。ここでRNPの疎水性コア内にはアミノ基を導入しているため、アミン塩基が触媒になってゾルゲル反応が進行するとともに形成したシリカへのアミノ基の配位によりRNPの物理架橋が起こり安定化することがポイントとなる。作製したsiRNPの②物理化学特性、③安全性試験、④吸着特性、⑤抗酸化試験、⑥動物評価を中心に検討を行った。siRNPからの未反応物の漏出や腹腔内からリンパ系を介しての血中取り込みを避けることが重要であり、ESR、ICP-MS等を利用して解析した。また吸着能の評価をin vivoで行った。さらには腹膜に生じる活性酸素種の消去に伴うサイトカイン産生、脂質過酸化、タンパクカルボニル化、8-OHdG定量により腹膜の酸化ストレスを評価するとともにNF-κBの活性化抑制効果を見積もった。このようにして新しいsiRNPによって被嚢性腹膜硬化症のメカニズムをin vivoで評価した。さらに後期ではヒドロキシアパタイトやポーラスシリカ等を利用した新しい融合材料の調製を行い、腹膜透析における老廃物吸着能の向上を達成した。このように本研究では安全でQOLの高い腹膜透析法の創出を行い、高い可能性を示すことが出来た。さらに作成したsiRNPは消化管障害の少ない経口DDS材料として評価し、高い有用性を示した。
