Outline of Annual Research Achievements

H27年度は、FUSによる転写抑制がその後のRNA代謝に、どのような影響を与えるか明らかにするため、RNA結合部位を網羅的に同定するCLIP-seq、RNAP II集積を解析するRNAPII ChIP-seq、転写開始点変化を明らかにするCAGE-seq、スプライシング変化を明らかにするdirectional mRNA-seq、転写終結点変化を明らかにするpolyA-seq、転写中のRNAを明らかにするnascent-seqの6種のhigh throughput sequencingのdataを用いて、それらの統合解析を行った。
Fus knockdown細胞を用いたCAGE-seq, RNA-seq, polyA-seq, CLIP-seq,の統合解析により、FUS-RNA結合部位周囲+-1000ntの範囲内に、選択的転写開始点が平均1個、選択的スプライス部位が平均1.5個存在していたのに対し、選択的転写終結点は、平均5個と顕著に存在していた。Fus knockdownにより、発現上昇する転写終結点が1033個、逆に発現低下する終結点が1977個みられ、FUSは転写終結を促進も抑制もできることが明らかとなった。さらにCLIP-seqとNascent-seq、RNAPII ChIP-seqの統合解析を行ったところ、Fus knockdownにより発現上昇した転写終結点では、その上流にFUS-RNA結合、RNAPIIうっ滞、新生RNAの減少が認められ、逆に発現低下する終結点では、下流にFUS-RNA結合、RNAPIIうっ滞、新生RNAの減少が認められた。RNAPIIうっ滞、新生RNAの減少は、転写抑制を意味する。転写抑制は転写終結に至る重要な過程の一つであり、以上の結果は、FUSが局所的な転写抑制により転写終結を部位特異的に制御している、ことを示唆した。
研究成果をまとめ、GENES & DEVELOPMENT誌に学術論文を、その概説をATLAS of Science誌に、さらにFUSによるRNA代謝についてのレビューをWiley Interdiscip Rev RNA誌に発表した。
